Research Interests:
1. Currently working on a project on K+ ion channels (Kir channels), relationship between endothelial stiffness and cellular cholesterol metabolism. Some of our findings with Primary endothelial cells from the lumen of femoral, coronary and carotid arteries from pigs fed on high and low cholesterol diet have shown that the Kir Current is suppressed in pigs fed on high cholesterol diet. The 86Rb+ flux assay, patch clamping also confirmed the suppression of kir current. MSH cells from undifferentiated Bone Marrow derived cells have shown that kir current is regulated by cholesterol. Ex-vivo culture of coronary arteries and transfecting the arteries with adeno and lenti-virsuses 2. Effect of Conjugated Fatty acid on the lipid metabolism”. Isolation and characterization of lipid rafts. Effect of conjugated fatty acid on the membrane fluidity. Synthesis of synthetic conjugated phospholipids and their interaction with cholesterol. Cell culture studies (Effect of SPF protein on Cholesterol metabolism in McARH 7777 cell line and HEPG2 Cells. Effect of Garlic and Garlic Compounds/ Tea (Black Tea and Green Tea)/ Tea Compounds and Policosanol on Cholesterol biosynthesis in cell culture. Study of various enzymes involved in cholesterol biosynthetic pathway and use of various inhibitors in cell culture to demonstrate effect of garlic Tea and Policosanol. 3. Physiological Role of Sphingomyelin in the metabolism of Plasma lipoproteins. Regulation of secretory phospholipase (V and X) by Ceramide and effect of cis and Trans sphingomyelin on plasma lipoproteins metabolism. General techniques used in Lipid biochemistry. 4. Enzymology: Standardization of enzyme assays, Enzyme/ Protein purification (using chromatographic techniques, Ion exchange. Affinity, gel filtration etc) and characterization. 5. Cloned and purified Human Supernatant Protein factor (SPF) and Rat Supernatant protein factor- like protein (SPF2), Squalene monooxygenase and P-450 reductase in E. coli, we have studied the regulation of Cholesterol synthesis by SPF and involvement of this entire enzyme. Some of the data has already been published in Journal of Biological Chemistry (see the reference section). 6. 86Rb+ flux assay for Ion Channels, Patch clamping for electrophysiology, Characterization of human proteins through their expression and purification in E. coli. Working knowledge of recombinant DNA techniques to clone the cDNAs and then express the encoded proteins using bacterial expression vectors. Purification of enzymes/protein using various techniques including affinity chromatography. Possess strong working knowledge in enzyme assays, ELISA methodology, electrophoresis, protein isolation and purification, cell biology and Western blotting, immuno-precipitation, RT-PCR techniques, and Radioactive- enzyme assays, human plasma analysis (separation of HDL, LDL, and VLDL by density gradient).
